ABSTRACT

Abstract The objective of this study was to evaluate the ability to degrade organic matter by edaphic macrofauna (worms), carbon/nitrogen (C/N) ratio and hydrogenation potential (pH) during the vermicomposting process in different organic residues. The treatments were constituted by organic residues of animal origin (bovine, ovine and equine manure) and vegetable (herb-checkmate and coffee drag), which were conditioned in plastic pots with a capacity of 10 liters, comprising five treatments in a completely randomized experimental design, with five replications. Were inoculated 150 earthworms of the species Eisenia foetida, into each plot. After 87 days, the evaluation of the multiplication of the earthworms was carried out, through its manual count and its cocoons. At the beginning and at the end of the experiment, the samples were submitted to analysis of humidity at 60 °C, pH, volumetric density, chemical analysis of macronutrients and C/N ratio. There was a dominance of worms and cocoons in the process of vermicomposting in the residues of ovine manure and herb-checkmate. The macronutrients (P, K and Mg) and C/N ratio were higher in the vegetal residues, while for N higher values were found in ovine manure and coffee drag treatments, and for Ca higher value among treatments was observed in the coffee drag treatment at the end and the lowest value at initiation. The results obtained in this study demonstrate the importance of the edaphic macrofauna to the vermicomposting process, since it allows more information about its influence on the continuity of soil organic matter decomposition processes.


Resumo Este estudo objetivou avaliar a capacidade de degradar a matéria orgânica pela macrofauna edáfica (minhocas), a relação carbono/nitrogênio (C/N) e o potencial hidrogeniônico (pH), durante o processo da vermicompostagem em diferentes resíduos orgânicos. Os tratamentos foram constituídos por resíduos orgânicos de origem animal (esterco bovino, ovino e equino) e vegetal (resíduo de erva-mate e borra-de-café), os quais foram acondicionados em vasos plásticos com capacidade de 10 litros, compondo cinco tratamentos em delineamento experimental inteiramente casualisado, com cinco repetições. Foram inoculadas 150 minhocas da espécie Eisenia foetida, em cada recipiente. Após 87 dias, foi realizada a avaliação da multiplicação das minhocas, através da sua contagem manual e seus casulos. Os resíduos foram submetidos, ao inicio e ao final do experimento, a análises de umidade a 60 °C, pH, densidade volumétrica, análise química de macronutrientes e relação C/N. Houve uma dominância de minhocas e casulos no processo da vermicompostagem nos resíduos de esterco ovino e erva-mate. Observou-se para os macronutrientes (P, K e Mg) e para a relação C/N uma maior quantidade nos resíduos vegetais, enquanto que para N valores maiores foram encontrados nos tratamentos esterco ovino e borra-de-café, e para Ca o maior valor entre os tratamentos foi observado no tratamento com borra de café ao final e o menor valor no inicio. Os resultados obtidos neste estudo demonstram a importância da macrofauna edáfica para o processo da vermicompostagem por possibilitar maiores informações sobre sua influência na continuidade dos processos de decomposição da matéria orgânica.

ABSTRACT

OBJECTIVE:To establish a method for the simultaneous determination of uracil,hypoxanthine,xanthine,uridine and adenosine in Eisenia foetida. METHODS:HPLC was performed on the column of Venusil XBP C18 with mobile phase of 50%methanol-0.01 mol/L potassium dihydrogen phosphate solution(gradient elution)at a flow rate of 1.0 ml/min. The detection wave-length was 254 nm with a column temperature at 30 ℃,and the injection volume was 10 μl. RESULTS:The linear range was 0.05-1.60 μg for uracil(r=0.9991),0.05-1.60 μg for hypoxanthine(r=0.9993),0.002-0.010 μg for xanthine(r=0.9991), 0.0075-0.24 μg for uridine(r=0.9999)and 0.0250-0.80 μg for adenosine(r=0.9990);RSDs of precision,stability and accuracy tests were lower than 3%;recoveries were 98.30%-100.76%(RSD=0.8%,n=9),98.68%-100.96%(RSD=0.8%,n=9), 95.16%-97.67%(RSD=0.9%,n=9),96.15%-99.57%(RSD=1.5%,n=9)and 96.39%-101.93%(RSD=1.8%,n=9),respective-ly. CONCLUSIONS:The method is simple and stable with good reproducibility,and can be used for the simultaneous determina-tion of uracil,hypoxanthine,xanthine,uridine and adenosine in E. foetida.

ABSTRACT

The earthworm Eisenia foetida (Annelida, Oligochaeta) is an excellent biosentinel model organism for assessing potential toxicological risks by xenobiotics in terrestrial ecosystems. Methamidophos is an organophosphorous insecticide widely used to control agricultural pests. Its main action is due to inhibition of the enzyme acetylcholinesterase. The aim of this study was to evaluate the toxicity of methamidophos on the morphology and male parameters in Eisenia foetida. After determining the LD50 for methamidophos (85.34 mg/kg soil), five groups were formed, including control subjects, and sprayed with tap water. Methamidophos treated groups were subjected to doses of 1/10, 1/6, 1/3 and 2/3 of the LD50. Each group was analyzed at day 1, 6, 14 and 30 post-treatment. The results show a significant decrease in the weight of all individuals treated with methamidophos, besides showing a coiled body in 100 percent of them. Sperm count showed a significant increase in the treated groups after 1, 6 and 14 days post exposure and a significant decrease by day 30. Using the technique of acridine orange, a significant increase in abnormal metachromatic sperm (red fluorescence) in treated animals was shown. Methamidophos alters the overall behavior in earthworms and the male reproductive parameters of Eisenia foetida possibly altering the structure and conformation of DNA.


La Lombriz de tierra, Eisenia foetida (Annelida, Oligochaeta), constituye un excelente modelo de organismo biocentinela para evaluar los riesgos tóxicos de xenobióticos, en ecosistemas terrestres. Metamidofos es un insecticida organofosforado de amplio uso para controlar plagas agrícolas. Su acción principal se debe a la inhibición de la enzima acetilcolinesterasa. El objetivo de este estudio fue evaluar la toxicidad de metamidofos sobre la morfología y parámetros espermáticos de Eisenia foetida. Una vez determinada la LD50 para metamidofos (85,34 mg/kg de tierra), se tomaron 5 grupos, entre ellos los individuos del grupo control, los cuales fueron asperjados con agua potable. Los grupos tratados con metamidofos fueron sometidos a dosis de 1/10, 1/6, 1/3 y 2/3 de la LD50. Cada grupo fue analizado al día 1, 6, 14, y 30 post tratamiento. Los resultados muestran una baja significativa en el peso de todos los individuos tratados con metamidofos, además de presentar enrrollamiento corporal, en el 100 por ciento de ellos. El recuento espermático presenta un aumento significativo en los grupos tratados a 1,6 y 14 días, para luego decaer significativamente el día 30. El uso de la técnica del naranja de acridina, mostró un aumento significativo de espermatozoides metacromáticos anormales en los animales tratados (fluorescencia de color rojo). Se concluye que Metamidofos altera el comportamiento general de la lombriz de tierra y los parámetros reproductivos espermáticos de Eisenia foetida posiblemente alterando la conformación y estructura del ADN.

ABSTRACT

AIM:To identify the optimal extraction of anti-asthma active protein from fresh Earthworm(Eisenia foetida).METHODS:Extraction time,extraction solvent and pH were selected as influencing factors.90 quinea pigs were randomly divided into 9 groups(control,montelukast,low temp,room temp,acidic buffer extract,alkaline buffer extract,aqueous extract,neutral buffer extract,powder earthworm group).Anti-asthma effciency was compared among groups.extraction time,saturation of ammonia sulfate buffer volume were selected as factors of orthogonal design.RESULTS:The optimal extraction for anti-asthma protein was at 4 ?C,adding 2 fold volume of pH 6.8 buffer phosphate,homogenating,standing and extracting for 3 hours,taking the supernataut and centifuging,salting out with 80% ammonia sulfate,suspecting with 20 mmol/L,pH6.8 buffer phosphate.So Earthworm protein was gained from the clear supernatant.CONCLUSION:The extraction technique may keep its original activity.
